产品概述

Cy7酪胺是美国AAT Bioquest生产的Cy系列产品,对于许多免疫组织化学(IHC)应用,传统的酶促扩增程序足以实现足够的抗原检测。但是,有几个因素限制了这些程序的敏感性和实用性。事实证明,酪胺信号放大(TSA)是一种特别通用且功能强大的酶扩增技术,具有更高的检测灵敏度。 TSA基于HRP在低浓度过氧化氢存在下将含有标记酪胺的底物转化为氧化的高反应性自由基的能力,该自由基可与HRP处或附近的酪氨酸残基共价结合。为了实现大IHC检测,酪胺用荧光团预标记。每个过氧化物酶标记物的多个酪胺底物转换所赋予的信号放大转化了对低丰度靶标的超灵敏检测以及使用较少量的抗体和杂交探针。在免疫组织化学应用中,源自TSA方法的灵敏度增强允许增加一级抗体稀释以减少非特异性背景信号,并且可以克服由次优固定程序或低水平靶表达引起的弱免疫标记。 Cy7酪胺含有明亮的Cy7,可以使用标准的Cy7滤波片组轻松检测到。百萤生物是AAT Bioquest的中国代理商,为您提供优质的Cy7酪胺。

实验方案

样品染色示例

概述

1.固定/透化/阻断细胞或组织
2.在封闭缓冲液中加入一抗
3.加入HRP偶联的二抗
4.准备酪胺工作溶液,在室温下在细胞或组织中施用5-10分钟

操作步骤

        在没有额外说明的情况下,所有未使用的储备溶液应分成一次性等分试样,并在制备后储存在-20°C,避免反复冻融循环。该方案适用于细胞和组织染色。

1.制备储存溶液

Tyramide原液(1000X)加入适量的DMSO,制成1-5mM的酪胺储备液。
注意:未使用的Tyramide原液可以在2-8℃下储存

2.制备工作溶液

Tyramide工作溶液(1X):
将1μLTyramide原液加入1 mL含有0.003%H2O2的缓冲液中。
注意:Tris Buffer,pH = 7.4时可用于获得佳性能。
注意:Tyramide工作溶液应立即使用。

3.细胞固定和透化

3.1在室温下用PBS中的3.7%甲醛或多聚甲醛固定细胞或组织20分钟。
3.2用PBS冲洗细胞或组织两次。
3.3在室温下用0.1%Triton X-100溶液使细胞透化1-5分钟。
3.4用PBS冲洗细胞或组织两次。
注意:组织固定,脱蜡和再水化,根据标准IHC方案对组织进行脱蜡和脱水。根据需要使用优选的特定溶液/方案进行抗原修复。

4.过氧化物酶标记

4.1任选:通过在过氧化物酶猝灭溶液(例如3%过氧化氢)中孵育细胞或组织样品10分钟来淬灭内源性过氧化物酶活性,在室温下用PBS冲洗两次。 
4.2可选:如果使用HRP偶联的链霉抗生物素蛋白,建议通过生物素阻断缓冲液阻断内源性生物素。
4.3用优选的封闭溶液(例如含有1%BSA的PBS)在4℃下封闭30分钟。
4.4取出封闭溶液,加入在推荐抗体稀释液中稀释的一抗在室温下60分钟或在4°C下过夜。
4.5用PBS洗涤三次,每次5分钟。
4.6向每个样品中加入100μL二抗-HRP工作溶液,在室温下孵育60分钟。
注意孵育时间和浓度可根据信号强度而变化。
4.7用PBS洗涤三次,每次5分钟。

5.酪胺标记

5.1为每个样品准备并应用100μL酪胺工作溶液,并在室温下孵育5-10分钟。
注意:如果您观察到非特异性信号,可以缩短酪胺的孵育时间。您应该在不同的孵育时间点使用阳性和阴性对照样品优化孵育期。或者您可以在工作溶液中使用较低浓度的酪胺。
5.2用PBS冲洗三次。

6.荧光成像

1.根据需要对细胞或组织样本进行计数。AAT提供了一系列细胞核复染试剂,如表1所列。按照试剂提供的说明进行操作。
2.使用具有抗褪色特性的安装介质安装盖玻片。
3.使用适当的过滤器组可视化来自酪胺标签的信号。
