BrdU 细胞增殖检测试剂是美国AAT生产的用于细胞增殖的产品,染料修饰的脱氧尿苷5'-三磷酸(例如氨基烯丙基dUTP)可用于通过常规酶促掺入方法(例如逆转录,切口平移,随机引物标记或PCR)产生含染料的DNA。该酶荧光标记方法广泛用于FISH探针和基于微阵列的实验。该TMR-dUTP缀合物可用作具有Spectrum Orange®过滤器组的橙色荧光颜色(Spectrum Orange®Vysis的商标)。百萤生物是AAT Bioquest的中国代理商,为您提供最优质的细胞增殖检测试剂/试剂盒。 

实验方案

用于染色细胞的样品方案

以下程序适用于大多数细胞类型。 生长培养基,细胞密度,其他细胞类型的存在和其他因素可能影响Brdu掺入。

 

1)制备1-10mM DMSO储备溶液。 如果储存在-20°C,DMSO储备液可以保存6个月。

2)通过离心沉淀细胞并将细胞重悬于完全生长培养基中。 培养物中的贴壁细胞可以在盖玻片上或细胞培养孔中原位染色。

3)使用5和20μM之间的浓度添加Brdu染色并孵育60分钟过夜作为指导。 最佳孵育时间取决于细胞类型和实验目标。

4)用70-80%酒精或酸 - 乙醇固定细胞20-30分钟。

5)用PBS洗涤(3次,每次2分钟)。

6)继续进行免疫细胞化学染色技术。
