几乎所有的质膜都具有跨膜电位,膜内侧相对于膜外侧通常为负电位。信号是通过在膜的某一点上打开或关闭离子通道,产生膜电位的局部变化而产生的。这种电场的变化可以迅速地受到膜中相邻或更远的离子通道的影响。这些离子通道由于电位的变化可以打开或关闭,从而再现信号。Cell Meter™荧光细胞电压检测试剂盒使用FRET对( VSB 405和VSR 555)来监测细胞膜电位的变化。亲脂性VSB 405主要位于脂质膜的外层,而VSR 555的定位对细胞膜电位敏感。在静息状态下,内层具有相对负电位,使得VSR 555主要位于靠近细胞膜外层,从而靠近蓝色荧光VSB405,导致有效的荧光从蓝色( VSB 405 )转移到红色( VSR 555 )。当细胞去极化时,VSR 555转移到细胞膜的内层,从而分离FRET对并破坏FRET。蓝色/红色荧光的比率与电池电压成正比。该方法可用于筛选调节离子通道的化合物。
