实验方案

实验方案

概述

1.添加 100 µL 核酸标准品或测试样品
2.添加 100 µL Helixyte Green ALL溶液
3.在室温下孵育 5-10 分钟
4.检测Ex/Em=490/525 nm 处的荧光强度

注:以下方案是使用 Helixyte Green All 定量总核酸含量的示例。 打开前让所有成分升温至室温。 没有关于 Helixyte Green All(总核酸染色剂)的致突变性或毒性的数据。 由于该试剂与核酸结合,因此应将其视为潜在的诱变剂并小心处理。 应特别小心处理 DMSO 原液,因为已知 DMSO 可促进有机分子进入组织。

 

溶液制备
核酸标准品
将 10 uL 100 ug/mL 核酸标准溶液(组分 C)加入 190 uL 检测缓冲液(组分 B)中,得到 5 ug/mL 标准溶液,然后进行 1:3 稀释,得到连续稀释的核酸标准溶液( NS1-NS7)。请使用AAT Bioquest连续稀释计算器以方便数据处理。

 

Helixyte Green ALL工作溶液
将 100 μL Helixyte Green All(组分 A)加入 10 mL 检测缓冲液(组分 B)中,制备 Helixyte Green All 工作溶液。 通过用箔纸覆盖或将其置于黑暗中来保护工作溶液免受光照。
注意:建议在塑料容器而不是玻璃容器中制备工作溶液,因为染料可能会吸附到玻璃表面。 为获得佳效果,该溶液应在稀释后几小时内使用。
注意:10 mL 的工作溶液对于一个 96 孔板就足够了。

1.根据表 1 和 2 中提供的布局准备核酸标准 (NS)、空白对照 (BL) 和测试样品 (TS)。对于 384 孔板,每孔使用 25 µL 试剂,而不是 100 µL。

2.将 100 µL Helixyte Green All 工作溶液加入核酸标准品、空白对照和测试样品的每个孔中,使测定体积为 200 µL/孔。 对于 384 孔板,将 25 µL Helixyte Green All 工作溶液添加到每个孔中,以获得 50 µL/孔的总体积。
3.在室温下孵育反应 5 到 10 分钟,避光。
4.使用 Ex/Em = 490/525 nm(Cutoff在 515 nm )处的荧光酶标仪检测荧光增加。
