产品介绍

聚组氨酸是掺入重组蛋白中流行的亲和标签之一。它可以插入N端或C端,并在多种宿主中表达。由于其体积小,聚组氨酸标签可作为蛋白质纯化和检测的理想工具。 HIS Lite™Cy3 Bis NTA-Ni和Cy5 Bis NTA-Ni复合物可对His标记的融合蛋白进行特异性和高灵敏度的检测。 Ni-NTA配合物早由Kapanidis等报道。对具有小交叉反应性的多组氨酸标签具有特异性。 Cy3和Cy5染料在通常可用的波长处显示强荧光信号,几乎没有猝灭。 Cy3 Bis NTA-Ni和Cy5 Bis NTA-Ni复合物可直接应用于SDS-PAGE凝胶或Western blot膜进行荧光成像。用Cy3 Bis NTA-Ni和Cy5 Bis NTA-Ni复合物进行检测所需的孵育时间少于蛋白质-抗体结合所需的孵育时间。由于Ni-NTA络合物直接与荧光团偶联,因此不需要二次反应。
