产品介绍

Hoechst 染料是一系列用于荧光显微镜标记 DNA 的荧光染料。因为这些荧光染料可以标记 DNA,所以它们也常用于可视化细胞核和线粒体。其中两种密切相关的双苯并咪唑衍生物常被使用:Hoechst 33258 和 Hoechst 33342。这两种染料都在大约 350 纳米的紫外线下激发,并在 461 纳米的发射峰处发出蓝色/青色的荧光。Hoechst 染料可以用于活细胞或固定细胞,通常用作另一种核酸染料 DAPI 的替代品。它们之间的主要区别是 Hoechst 33342 添加的乙基使其更亲脂,因此更能穿透完整的细胞膜。在某些应用中,Hoechst 33258 的渗透性较低。这些染料还可以通过绘制标准的发射量与内容曲线来检测样本 DNA 的含量。

实验方案

染色细胞分析方案       

        以下程序适用于大多数细胞类型。 生长培养基,细胞密度,细胞类型和其他因素可能影响染色。 玻璃器皿上的残留洗涤剂也可能影响许多生物染色,导致在没有细胞存在的溶液中出现明亮染色的物质。

        通过离心沉淀细胞并将细胞重悬于缓冲盐溶液或培养基中,经过测试染料的佳结合pH为7.4。培养物中的贴壁细胞可以在盖玻片上或细胞培养孔中原位染色。Hoechst的染色浓度为0.5-5uM,并将其孵育15至60分钟。在初的实验中,好在整个建议范围内尝试几种染料浓度,以确定产生佳染色的浓度。
