相较于其他商业过氧化氢溶液,AAT Bioquest制备的过氧化氢溶液具有更高的稳定性。经精确标定,可确保过氧化物酶相关检测结果具有更优异的重复性。
重要提示
本方案适用于大多数过氧化物酶-过氧化氢检测体系。初步实验建议在推荐浓度范围内测试多个过氧化氢溶液浓度,以确定最佳反应条件。
储备液配制
除非另有说明,所有储备液均应分装为单次使用量,制备后于-20°C保存,避免反复冻融。
过氧化物酶反应工作液:
配制含适宜过氧化氢底物浓度及50-500 µM过氧化氢的过氧化物酶反应体系。
实验方案
1.向待测孔中等体积加入过氧化物酶工作液、标准品及含过氧化物酶的样品
2.室温避光孵育15-30分钟
3.使用相应仪器检测荧光或吸光度信号
操作方案
1.储备溶液配制
所有未使用的储备溶液应分成一次性等分试样,并在制备后储存在-20°C。避免反复冻融循环。
1.过氧化物酶反应混合物:
用适当的过氧化氢底物浓度和50至500μM过氧化氢制备过氧化物酶反应混合物。
样品实验方案
1.加入等体积的过氧化物酶工作溶液和过氧化物酶标准品和含过氧化物酶的样品。
2.在室温下孵育反应15至30分钟,避光。
3.用适当的仪器监测荧光或吸光度。
