产品介绍

细胞非渗透性探针Mag-Fluo-4 AM是Fluo-4的衍生物,其镁离子解离常数(Kd)为4.7 mM,钙离子解离常数(Kd)为22 μM。这一特性使其兼具双重功能:可作为细胞内镁离子指示剂,也可作为低亲和力钙离子指示剂,该探针能够精确追踪骨骼肌细胞中空间平均游离钙离子瞬变的动力学过程,为研究骨骼肌钙信号转导提供可靠的动力学数据。

实验方案示例

储备液配制

所有未使用的储备液应分装为单次用量,配制后保存于-20°C,避免反复冻融

Mag-Fluo-4 AM储备液

使用高质量无水DMSO配制2-5 mM的Mag-Fluo-4 AM储备液。

 

工作液配制

Mag-Fluo-4 AM工作液

实验当天,将Mag-Fluo-4 AM溶解于DMSO或解冻储备液至室温。

选择适当缓冲液(如Hanks和Hepes缓冲液),加入0.04% Pluronic® F-127,配制2-20 µM Mag-Fluo-4 AM工作液。多数细胞系推荐使用4-5 μM终浓度,具体浓度需根据实验条件优化

注意事项:1.Pluronic® F-127可提高Mag-Fluo-4 AM的水溶性,百萤生物提供多种Pluronic® F-127溶液

                  2.若细胞存在有机阴离子转运体,可在工作液中加入1-2 mM丙磺舒(孔内终浓度0.5-1 mM)以减少探针泄漏,百萤生物提供多种ReadiUse™丙磺舒产品

 

操作步骤

1.将细胞在生长培养基中培养过夜

2.次日向细胞板中加入1X Mag-Fluo-4 AM工作液

注:若待测化合物与血清有相互作用,加载前更换为新鲜HHBS缓冲液

3.孵育条件:37°C细胞培养箱中孵育30-60分钟

注:某些细胞系可延长孵育时间至1小时以上以提高信号强度

4.用HHBS或含阴离子转运抑制剂(如1 mM丙磺舒)的缓冲液替换工作液

5.加入刺激剂后,立即使用以下设备检测荧光信号:

配备FITC滤光片的荧光显微镜

荧光酶标仪(如FDSS、FLIPR或FlexStation)检测参数:Ex/Em = 490/525 nm,截止波长515 nm
