单氯二胺 (mBCl) 在与硫醇基团反应前基本无荧光。它能与多种低分子量硫醇迅速反应，包括半胱氨酸、谷胱甘肽、N-乙酰半胱氨酸、巯基嘌呤、肽类及血浆硫醇。mBCl与谷胱甘肽形成的加合物，其最大吸收/发射波长分别约为394/490nm。
