丙磺舒是一种位于细胞膜上的有机阴离子转运蛋白抑制剂。这类转运蛋白常会将荧光指示剂从细胞内排出,从而导致染料滞留性差。该现象通常在需要染料指示剂良好滞留于细胞内的实验中引起高背景荧光。使用丙磺舒抑制转运蛋白活性,从而减少细胞内染料指示剂的泄漏,是降低钙检测实验中荧光背景的常用方法。

常用的丙磺舒游离酸形式由于在中性水中溶解度差,需使用1 M NaOH进行溶解。AAT Bioquest提供便捷的ReadiUse™水溶性与热稳定性丙磺舒产品,有粉末、溶液或片剂等多种规格。这些产品使用方便,且在相同浓度下与游离酸形式效果相当。每瓶试剂用90 mL缓冲液稀释后,可获得终浓度为2.5 mM的丙磺舒溶液,可直接用于实验。

实验方案

操作步骤

1.解冻1瓶#20062在室温下使用。

2.制作2.5 mM ReadiUse 丙磺舒缓释缓冲液:将9 ml HHBS(1X Hank's含20 mM Hepes缓冲液,pH 7.0)加入1 ml ReadiUse 丙磺舒缓释25mM稳定水溶液。

注1:10 mL的2.5 mM Readiuse 丙磺舒缓冲液足以用于1个板。 分装并保存未使用的Readiuse 丙磺舒25 mM稳定的水溶液,温度<-20℃,避光。 避免重复冻融循环。

注2:2.5 mM Readiuse 丙磺舒缓冲液含有0.02%的Pluronic®F-127,因此无需在染料加载溶液中添加额外的Pluronic®F-127。

3.为一个细胞板制备上样溶液:添加DMSO重构的荧光钙染料(例如Indo-1 AM,Fura-2 AM,Fluo-3 AM,Fluo-4 AM,Fluo-8 AM,Rhod-2 AM和 Rhod-4 AM)加入2.5 mM ReadiUse 丙磺舒缓冲液中,并充分混合。 该工作溶液在室温下稳定至少2小时。
